About finding a spot (clue scrolls)

Can the spot be on the 2nd ground level? And can you detect it from the 1st ground level? I have to find a place in Varrock, and I am rather sure that I have searched every inch, without any result.

It can't. The only exception is the upper floor in Rimmington/Catherby dungeon.

Check city's nearby areas (up to champions' guild, GE, the large pub south-east of Varrock etc)

Try outside varok wall at sawmille near at yew tree. And try grand exhange around wall.
